Description
StockOptions ADA buffer kit is a preformulated, sterile filtered set of titrated buffer stocks. The StockOptions buffer stock reagents are supplied as 1.0 M stock solutions in 10 milliliter volumes. Each StockOptions ADA buffer reagent is carefully titrated using Sodium hydroxide. StockOptions ADA is comprised of 20 unique reagents covering the pH range of 5.6 to 7.5 in 0.1 pH unit increments.

Description
Grid Screen™ MPD is a preformulated reagent kit designed to provide a rapid screening method for the crystallization of biological macromolecules.
The screen is simple and practical for finding initial crystallization conditions as well as determining the solubility of a macromolecule in (+/-)-2-Methyl-2,4-pentanediol (MPD) between pH 4.0 and 9.0.
Grid Screen MPD evaluates (+/-)-2-Methyl-2,4-pentanediol (MPD) at four concentrations (10, 20, 40, 65% v/v) versus six pH levels (4, 5, 6, 7, 8, 9).
Grid Screen MPD consists of 24 unique reagents, each supplied as 10 milliliter in sterile, screw cap tubes. Ready-to-use reagents are sterile filtered and formulated with ultra-pure Type 1 water, using the highest purity polyol and buffers. Individual reagents are available through the Hampton Research Custom Shop.

Description
24 well plate with sealant for hanging drop vapor diffusion crystallization with a 128 mm x 85 mm microplate footprint. Stackable, optically clear plastic, 24 well plates with raised covers (to allow room for cover slides) and flat bottoms for exceptional optics. Raised, wide rings around each reservoir (well) minimize cross contamination and allow each well to be individually sealed with 18 mm diameter circle cover slides. Plates are individually wrapped and supplied with applied sealant.
The microplate footprint is space-saving and compatible with automated liquid handling systems.

Parabar 10312 is a viscous cryoprotectant for both small and large molecule crystallography.1
Mildly air-unstable, small molecule compounds can be coated with Parabar under an inert atmosphere. The Parabar protected crystal sample can be cryocooled in a chilled nitrogen gas stream. Parabar has also been used successfully as a cryoprotectant for biological macromolecule crystals. Used during cryocrystallography to displace and reduce the amount of water (mother liquor, reagent) on the crystal after the crystal is mounted in a cryoloop. Coating the crystal with Parabar can minimize evaporation from the crystal and reduce exposure and slow diffusion of air (oxygen) to the crystal.
HR2-643 Parabar 10312 is no longer available and has been discontinued effective May 11, 2021.
HR2-862 Parabar 10312 is supplied as 5 ml in an amber glass vial with glass applicator rod in screw cap lid.
The viscosity of Parabar can be reduced to improve handling with fragile needle and blade crystals by mixing 50:50 with Paraffin Oil.
Parabar is a blend of polyisobutylenes with a Staudinger molecular weight of about 20,000 and contains about 20% polymer.
Refractive index 1.48487 at 20°C.
Kinematic viscosity 641 cSt at 100°C. Kinematic viscosity ratio 2.05 at 100°C.
Clear to slightly hazy is appearance.

Description
Crystal Clear Quartz capillaries that are extremely thin walled (approximately 10 micron wall thickness). The length of the capillary has a well defined diameter, with one end having a funnel shape and the other end closed. Quartz capillaries have a wall thickness of 0.01 mm and an overall length of 80 mm +/-5 mm. Quartz capillaries are available in a wide range of outside diameters from 0.1 mm to 5.0 mm. They are designed to mount, hold, and store small molecule and biological macromolecular crystals for x-ray data collection. Capillaries can also be used for crystal density measurements and crystal growth experiments. The capillaries can be sealed tightly against moisture and gases using wax, epoxy, or other sealing materials.
In determining what glass or quartz capillary is right for you, please refer to the “Linear Absorption Coefficient µ cm-1” table. This table indicates the amount of radiation that is absorbed by the capillary during x-ray data collection.
For 0.1 mm to 2.5 mm capillaries the open end capillary tube base size is 3.0 +/- 0.2 mm OD x 0.2 +/- 0.15 mm Wall thickness. For 3.0 and 3.5 mm capillaries the open end capillary base size is about 4.4 mm OD x 0.25 mm Wall thickness. For 4.0 and 5.0 mm capillaries the open end capillary based size is about 6.0 x 0.25 mm.
The Diameter is measured about 30 to 40 mm from the closed end (measuring instrument: LaserMicrometer LS 7500) The tolerances are as follows.

Description
Individual reagents from the Proti-Ace and Proti-Ace 2 kits. Each protease is supplied in a stable, lyophilized format in an optimized digest buffer. Simply add water when ready to use.
The unique freeze dried formulation of the Proti-Ace kit offers a much improved protease stability compared to liquid protease formulations. Each tube contains 0.1 mg of lyophilized enzyme and digest buffer as shown below. Add 100 microliters of deionized water (Type 1+) to each tube of lyophilized content to create a 1 mg/ml protease solution in digest buffer. Proti-Ace Dilution buffer is supplied as a ready to use solution.

Description
The Sodium malonate offered by Hampton Research is Malonic acid (CAS Number 141-82-2) titrated to pH 4.0-8.0 using Sodium hydroxide as described in the publication “A comparison of salts for the crystallization of macromolecules. Alexander McPherson. Protein Science (2001), 10:418-422.”. In this same publication, Sodium malonate was reported to be more successful than any other salt, resulting in the crystallization of 19 of the 23 macromolecules, almost twice as effective as the next most successful salt, which was a draw between sodium acetate, sodium tartrate, sodium formate, and ammonium sulfate. The high success rate of Sodium malonate in producing crystals was even more impressive when an overall unique success rate with individual macromolecules was considered.
The use of Sodium malonate in place of Ammonium sulfate has been reported in one instance to improve the reproducibility of crystals and improve resistance to physicochemical shocks. Sodium malonate also acted as a good cryoprotectant (Xing and Xu 2003).
One may combine the four different sodium malonate stocks to create a pH versus sodium malonate concentration grid for screening or optimization. Refer to the pH/Dilution Table for Sodium malonate.
Based on the structure of malonic acid titrated with sodium hydroxide, there are no chiral carbons so it is not characterized as having enantiomeric structures, such as D or L.

Doxycycline diets
Envigo Teklad diets makes a variety of doxycycline containing diets for your Tet regulated systems. TD.01306 with doxycycline hyclate added at 625 mg/kg to Teklad global 2018 rodent diet is stocked and can typically ship within 1-2 days (non-irradiated) or 1-2 weeks (irradiated). Considerations when choosing a base diet
- Teklad Global Rodent Diets® like 2018 do not contain alfalfa and are thus ideal if your work requires an alfalfa-free diet for fluorescent imaging, If further imaging clarity is needed, we can make purified diets with doxycycline.
- Teklad global 2018 (base diet in examples above) contains a moderate amount of soybean meal, a source of phytoestrogens. The table below includes several options of minimal phytoestrogen diets with the most common doxycycline concentration of 625 mg/kg diet. For other concentrations contact us. 1 Doxycycline hyclate is ~87% doxycycline
Advantages of diet delivery of doxycycline
- Typically the same doxycycline concentration can be used in diet as in water. Lower concentrations of doxycycline may be adequate with 625 mg/kg diet being the most common concentration.
- Diet offers protection from light. Water may have to be given in dark-colored or foil-wrapped bottles
- Diet is typically changed once per week. Water may need to be changed every few days
- Diet delivery may also reduce risk of dehydration and preclude the need to give with sucrose (Cawthorne et. al. 2007)
Key planning information
- Minimum order quantity is 3 kg, sufficient for feeding ~20 mice for one month
- Store diet refrigerated and plan to use within six months
- Typical lead time is two weeks (four weeks if irradiated)
- TD.01306 typically ships within 2 days non-irradiated or 2 weeks with irradiation
- Irradiation (20 – 50 kGy) is optional, and must be requested at time of order
Safe handling of doxycycline diets
- Doxycycline is a tetracycline class antibiotic.
- Usual dose for therapeutic effects in humans is 100-200 mg/day.
- Doxycycline diets for rodents typically contain ~2 mg/pellet.
- Accidental doxycycline exposure can be minimized by using typical lab precautions of lab coat, gloves, and mask when handling the diet.
- Your chemical safety department should be contacted for additional institution specific guidelines for handling and disposal of doxycycline containing diets.

Description
To screen pH as a crystallization variable using Tacsimate one can mix together different ratios of different pH Tacsimate solutions. Refer to the pdf “Mixing Tacsimate Stocks to Screen pH” on this page. Or, one can add a buffer to the crystallizaiton reagent containing Tacsimate. Recommended final buffer concentration in the reagent well is 0.05 to 0.1 M.

Description
Siliconized glass coverslips allow a droplet to be suspended in a position which provides near optimal conditions for vapor diffusion with the surrounding reservoir solution. These siliconized glass cover slides provide a consistent, high quality finish for crystallization experiments. The hydrophobic surface produces a drop which “stands well” and does not flatten on the glass. The siliconized surface prevents the adhesion of crystals and precipitate onto the glass surface. Use vacuum grease or oil to seal cover slide to plate. Available in 18 or 22 mm diameter circles.

Description
SaltRx was developed by Hampton Research as a primary and secondary, salt and pH based crystallization screen for biological macromolecules.
Salt is the only primary crystallization reagent (precipitant) utilized. Based on a design of 96 conditions, the screen evaluates a broad portfolio of crystallization salts of varying concentration and pH. The selection, concentration, and pH of the salts were determined by data mining the BMCD1 and other crystallization databases, crystallization reports in the literature, and internal crystallization trials performed at Hampton Research. Based on this analysis, up to 35% of protein crystallizations involve salt as the primary crystallization reagent. SaltRx can be used as a primary crystallization screen when salt, ionic strength and pH are desired or suspected as appropriate crystallization variables. It is also useful as a secondary screen when salt only reagents/conditions from screens such as Index™, Crystal Screen™, and Grid Screen™ produce crystals and when further screening for additional salt conditions or optimization is desired. Since this screen does not contain volatile organics, it is compatible with microbatch, vapor diffusion, liquid and gel diffusion crystallization methods. SaltRx may also be used for dialysis crystallization in conjunction with Dialysis Buttons™.
The original SaltRx kit (HR2-108) contained 96 unique reagents, 10 ml each. This kit has been discontinued and converted into two 48 reagent based kits, SaltRx 1 and SaltRx 2, available separately, which when used together are identical to the original SaltRx.
SaltRx 1 contains 48 unique reagents, 10 ml each. SaltRx 1 contains reagents 1-48 of the original SaltRx 96 reagent kit.
Salt Rx 2 contains 48 unique reagents, 10 ml each. SaltRx 2 contains reagent 49-96 of the original SaltRx 96 reagent kit.
SaltRx HT contains the 96 SaltRx 1 and SaltRx 2 reagents in a single Deep Well block format.
Ready-to-use reagents are sterile filtered and formulated with ultra-pure Type 1 water, using the highest purity salts, polymers, organics and buffers. Individual reagents are available through the Hampton Research Custom Shop.

Description
StockOptions Buffer kits are reagent toolboxes for the macromolecular crystal grower. Stock-Options reagent kits offer precisely formulated, high quality crystallization grade reagent stocks in convenient, cost-effective kits. The chemicals utilized in StockOptions kits are the same crystallization grade, ultra-high purity chemicals utilized in the Hampton Research kits. StockOptions reagents are carefully formulated under strict quality standards to ensure reliable performance and lot-to-lot consistency.
Preformulated stocks remove calculation, measurement, and formulation errors. No more second guessing how the reagent was formulated, what specific chemical was used, when it was made, or how to precisely reproduce that reagent when it is gone and more reagent is required for additional setups.
StockOptions™ Sodium Citrate buffer kit is a preformulated, sterile filtered set of titrated buffer stocks. The StockOptions buffer stock reagents are supplied as 1.0 M stock solutions in 10 milliliter volumes. Each StockOptions Sodium Citrate buffer reagent is carefully titrated using hydrochloric acid. StockOptions Sodium Citrate is comprised of 24 unique reagents covering the pH range of 4.2 – 6.5 in 0.1 pH unit increments.
Formulated using Sodium citrate tribasic dihydrate and HCl.
pKa1 = 3.1 pKa2 = 4.8 pKa3 = 6.4
Each StockOptions Sodium Citrate Buffer kit reagent is available separately as a 185 milliliter volume through the Custom Shop.

Description
The Cover Slide Vacuum Gadget™ allows one to pick up a single cover slide, invert the slide and gadget, rest the gadget on the bench so the slide rests as if on a pedestal, pipet the drop onto the slide, then use the gadget to place and release the slide onto the crystallization plate. The Gadget is a hand held vacuum bulb with a single Buna-N vacuum cup. Squeeze the bulb, press the vacuum cup to the slide, ease the squeeze on the bulb to create a vacuum and the slide is held in place at the end of the Gadget. When ready to position and place the slide give the Gadget a gentle squeeze to release the vacuum. Single bulb with 3/8″ (9.53 mm) diameter cup.
