Description

StockOptions Buffer kits are reagent toolboxes for the macromolecular crystal grower. Stock-Options reagent kits offer precisely formulated, high quality crystallization grade reagent stocks in convenient, cost-effective kits. The chemicals utilized in StockOptions kits are the same crystallization grade, ultra-high purity chemicals utilized in the Hampton Research kits. StockOptions reagents are carefully formulated under strict quality standards to ensure reliable performance and lot-to-lot consistency.

Preformulated stocks remove calculation, measurement, and formulation errors. No more second guessing how the reagent was formulated, what specific chemical was used, when it was made, or how to precisely reproduce that reagent when it is gone and more reagent is required for additional setups.

Rabbit, swine and other species diets

The most common custom rabbit diets are those with a standard diet base and added cholesterol. Typically, the added cholesterol ranges from 0.25% to two percent. Additional fat can also be a feature of these formulas. To preserve pellet quality we recommend no more than six percent additional fat. Added fat is most often supplied as peanut oil, corn oil, coconut oil, or a combination. We recommend the Teklad base diets 2030 or 2031.

Rabbits do not readily consume purified diet. When specific nutrient manipulations are necessary, hybrid diets or at least some alfalfa meal inclusion is recommended. Contact us for formula examples or to discuss your specific needs.

Most often, customized diets for swine feature natural ingredients with added fat and cholesterol. Here are three variations on this theme:

  • TD.92033 Swine diet (15% lard, 1.2% cholesterol)
  • TD.92052 Swine diet (15% lard, 1.5% cholesterol)
  • TD.93296 Swine diet (15% lard, two perecent cholesterol)

We recommend these diets in meal/powder form, as pellets are not very durable. Other natural ingredient and purified diets with various nutrient manipulations are available. Contact us for further information.

The most common custom Guinea pig diets are those with a standard diet base (such as Teklad 2040 or 2041) and added cholesterol.

Guinea pigs do not readily consume purified diets, so when specific nutrient manipulations are necessary, hybrid diets (mixture of refined and natural ingredients) are recommended. At the very least it is advisable to retain some alfalfa meal in the diet. Contact us for further information.

Most times, diets that are developed for mice and rats are suitable for hamsters and gerbils, and vice versa. We may advise increasing specific nutrients in a purified diet for hamsters and gerbils. Contact us for further information.

Depending on the focus of your research, customized diet for non-human primates can contain natural ingredients, refined ingredients, or a mixture of both. Custom diets for primates often feature adjustments in fat type, fat level, and cholesterol. Contact us for further information.

Description

24 well plate for hanging drop vapor diffusion crystallization with a microplate footprint. Stackable, optically clear plastic, 24 well plates with raised covers (to allow room for cover slides) and flat bottoms for exceptional optics. Raised, wide rings around each reservoir (well) minimize cross contamination and allow each well to be individually sealed with 18 mm diameter circle cover slides. Plates are individually wrapped and supplied without sealant.

The microplate footprint is space-saving and compatible with automated liquid handling systems.

Description

Grid Screen™ PEG/LiCl is a preformulated reagent kit designed to provide a rapid screening method for the crystallization of biological macromolecules.

The screen is simple and practical for finding initial crystallization conditions as well as determining the solubility of a macromolecule in PEG and Lithium chloride between pH 4.0 and 9.0.

Grid Screen PEG/LiCl evaluates Polyethylene glycol 6,000 at four concentrations (0, 10, 20, 30 %w/v) in the presence of 1.0 M Lithium chloride versus six pH levels (4, 5, 6, 7, 8, 9).

Grid Screen PEG/LiCl consists of 24 unique reagents, each supplied as 10 milliliter in sterile, screw cap tubes. Ready-to-use reagents are sterile filtered and formulated with ultra-pure Type 1 water, using the highest purity polymer, salt, and buffers. Individual reagents are available through the Hampton Research Custom Shop.

  • NDSB 256 4T is a non-detergent sulfobetaine (NDSB) that prevents protein aggregation and facilitates protein folding by interacting with the early folding intermediates. Reported to improve the in vitro renaturation efficiency of reduced hen egg lysozyme (60% enzymatic activity at 600 mM NDSB-256-4T) and chemically unfolded tryptophan synthase ß2 subunit (100% enzymatic activity at 1 M NDSB-256-4T). 

Description

The NDSB are a group of zwitterionic compounds that can reduce aggregation and aid in refolding proteins found in inclusion bodies and bacterial expression systems.

Non detergent sulfobetaines have a sulfobetaine hydrophilic group and a short hydrophobic group that cannot aggregate to form micelles, therefore NDSB’s are not considered detergents. NDSB increase the extraction yield (up to 30%) of membrane, nuclear and cyto-skeletal associated proteins. The short hydrophobic groups combined with the charge neutralization of the sulfobetaine group results in higher yields of membrane proteins. NDSB have been used in refolding and renaturation of chemically and thermally denatured proteins.

Typical useful NDSB concentration in protein sample is 0.5-1.0 M.

Description

In the dialysis method, the sample in question is separated from the “precipitant” by a semi-permeable membrane which allows small reagent molecules to pass but prevents biological macromolecules from crossing the membrane.1-3

Dialysis Buttons are injection molded from polystyrene, and are the size of a small button. The sample is placed in this chamber so as to create a slight dome of liquid at the top of the button. A dialysis membrane is placed over the top of the button/sample and is held in place with an O-ring. The O-ring is held in place by a groove in the Dialysis Button. Dialysis Buttons are supplied with O-rings. The 5 µl to 100 µl sized dialysis buttons have an approximate overall height of 6 mm, an approximate outside diameter of 10 mm without an O-ring in place, and an approximate diameter of 12 mm with an O-ring in place. The 200 µl and 350 µl sized dialysis buttons have an approximate overall height of 8 mm, an approximate outside diameter of 15 mm without an O-ring in place, and an approximate diameter of 17 mm with an O-ring in place. Each package of dialysis buttons includes a plastic golf tee shaped applicator to assist with applying a dialysis membrane to the button and securing with the supplied o-ring.

Description

Individual reagents from the Proti-Ace and Proti-Ace 2 kits. Each protease is supplied in a stable, lyophilized format in an optimized digest buffer. Simply add water when ready to use.

The unique freeze dried formulation of the Proti-Ace kit offers a much improved protease stability compared to liquid protease formulations. Each tube contains 0.1 mg of lyophilized enzyme and digest buffer as shown below. Add 100 microliters of deionized water (Type 1+) to each tube of lyophilized content to create a 1 mg/ml protease solution in digest buffer. Proti-Ace Dilution buffer is supplied as a ready to use solution.

• 1 mg/ml a-Chymotrypsin, 1 mM Hydrochloric acid, 2 mM Calcium chloride dihydrate
Proti-Ace Reagent 2* (Qty 3):
• 1 mg/ml Trypsin, 1 mM Hydrochloric acid, 2 mM Calcium chloride dihydrate
Proti-Ace Reagent 3* (Qty 3):
• 1 mg/ml Elastase, 200 mM Tris pH 8.0
Proti-Ace Reagent 4* (Qty 3):
• 1 mg/ml Papain, 2 mM EDTA, 2 mM TCEP hydrochloride, 2 mM L-Cysteine

Description

The CrystalCap ALS HT is a magnetic sample mount (also known as a cap, pin or goniometer base) designed for cryocrystallography systems that accept ALS style sample mounts. The CrystalCap ALS HT attaches to a magnetic CrystalCap Vial and magnetic goniometer base. The tip of the CrystalCap accepts a Mounted CryoLoop™.

The CrystalCap Vial is a 1.8 ml cryo vial featuring two small vents. A ring magnet is molded into the open end of the vial so that when the cap is positioned in the vial, the ring magnet holds the cap on the vial during cryogenic storage. The HR4-904 CrystalCap Vial does not have a magnet on the bottom of the vial.

The ledge free conical shape of the CrystalCap ALS HT is compatible with ALS style grippers, auto mounters and sample handlers.

The CrystalCap ALS HT features a two dimensional (2D) alphanumeric 16 x 16 data matrix code on the underside of the cap. Each cap is also color coded for CryoLoop size.

Note: CrystalCap ALS HT with Mounted CryoLoops are sold without vials. Vials HR4-904 sold separately.

Description

Low melting (LM) agaroses are the result of a derivatization process by organic synthesis. Essentially, the process generates methoxylate groups from the basic agarose structure. The main properties of these agaroses are their low melting and gelling temperatures when compared with standard agaroses. LM agaroses have lower gel strength than standard agaroses, yet they can be handled easily. LM agaroses have higher clarity (gel transparency) than gels of standard agaroses. LM agaroses have great sieving capacity. The gelling temperature of LM agaroses is 24 to 28°C.

Agarose is a neutral polysaccharide extracted from the cellular walls of Rhodophyceae algae belonging to the genera Gelidium, Gelidiella, Pterocladia, Gracilaria, and Ahnfeltia, also known as agarophyte seaweed. The structure of the polysaccharide is that of a galactan, formed by linking agarobioses by links 1-3, 1-4. This chemical structure gives agaroses the capacity to form strong gels even at low temperatures. The gels have a macroreticular structure with a very open mesh which can be adjusted simply by varying the concentration of the agarose. The macroreticule structure of the agarose gel is formed by hydrogen bonds, which makes the gel reversible, transforming the gel into a solution by heating. The hysteresis (difference between gelling and melting temperature) is greater than any other hydrocolloid. The absence of ionic groups makes the gel a neutral structure. With no interaction, macromolecules can migrate through the gel mesh, making the gel an efficient sieve for biological macromolecules.

The LM Agarose offered by Hampton Research is 100% pure, does not contain any additives, does not contain ligation inhibitors, and is free of DNAses and RNAses. Hampton Research LM agarose is clearer than other agaroses and also has a higher gel strength.

Description

The PCT Pre-Crystallization Test is used to determine the appropriate protein concentration for crystallization screening. Sample concentration is a significant crystallization variable. Samples too concentrated can result in amorphous precipitate, while samples too dilute can result in clear drops. Precipitate and clear drops are typical crystallization screen results for reagent conditions which do not promote crystallization and are part of every crystallization screen. However, by optimizing protein concentration for the screen, the number of clear and precipitate results can often be reduced, which in turn results in more efficient sample utilization while at the same time enhancing the chances for crystallization. PCT can minimize or prevent situations where a screen results in an over abundance of precipitate or clear drops.

The PCT kit contains 4 unique, preformulated, sterile filtered reagents used to evaluate protein concentration for crystallization screening. Initially, the sample protein is mixed with two of the reagents to determine if the protein concentration is appropriate for crystallization screening. If the protein is very sensitive to salt and polymer concentration, based on initial PCT results, the protein may be evaluated using a second set of PCT reagents. PCT results will then provide insight to either the appropriate sample concentration or indicate that other diagnostic testing such as native gel electrophoresis or dynamic light scattering should be performed to demonstrate sample homogeneity appropriate for crystallization.
