Rodent diets

As a researcher, you want to have as much control as possible over what goes into your study animals. At Envigo, we understand that. That is why we developed Teklad Global Rodent Diets® .
Teklad Global Rodent Diets® are a special integrated range of vegetarian laboratory rodent diets developed to be nutritionally complete for various life stages from breeding through long-term maintenance. Global rodent laboratory diets contain levels of protein, energy, vitamins and minerals that are more appropriate to the needs of modern biomedical research studies.
Furthermore, in global rodent diets particular attention has been placed on avoiding, as far as practical, ingredients that are reported to have adverse confounding effects on experimental results. This has resulted in a range of Teklad rodent laboratory diets that contain:
- No fish meal
- No meat meals or meat by-products
- No alfalfa meal
- No soybean meal or reduced levels
- No animal fat
By excluding animal by-products, the presence of nitrosamines (a potential carcinogen) is avoided. Exclusion of alfalfa meal reduces chlorophyll, improving optical imaging clarity. Reduction or removal of soybean meal, together with elimination of alfalfa meal, minimizes levels of naturally-occurring phytoestrogens. Traditional rodent diets were formulated decades ago based on understanding of rodent nutrition, ingredients, and diet manufacturing at the time. While traditional diets will supply the known nutrient needs of your laboratory animals, we recommend you consider the use of a diet from our newer global diet line for your modern research needs.
Teklad Global Rodent Diets® are modern formulas designed to reduce research variables. Specifically, these diets contain more appropriate nutrient levels, and limit or exclude ingredients that are reported to have effects on a wide variety of research endpoints.
- Lower, more appropriate protein levels can improve survival and reduce morbidity
- Vegetarian with no nitrosamines (a potential carcinogen)
- Formulated to exclude alfalfa meal, greatly improving fluorescent imaging clarity
- Formulated to exclude or lower soybean meal, thus minimizing the presence of isoflavones, the primary type of phytoestrogen found in lab animal diets
- Extruded rodent diets dramatically reduce clumping and hardness after autoclaving (2018SX , 2019S , 2020SX ), and in general result in less diet waste and cleaner cages
Teklad rodent diets are natural-ingredient diets specifically formulated to provide the proper balance of all known nutrients considered essential for the growth, maintenance, and reproduction of rats, mice, gerbils and hamsters. These diets conform to the nutrient requirements for rodents established by the National Research Council (1995).
Teklad rodent diets provide uniform nutrition. They are fixed-formula diets designed to minimize the nutrient variances which otherwise could occur if the ingredient composition of a diet were altered from one batch to the next.
Protein is supplied primarily by plant sources. Supplemental amino acids are added to provide the proper amount and balance of essential amino acids. All rodent diets are fortified with vitamins and minerals to help support the regulation of body fluids and the proper functioning of body systems to ensure the adequate growth, maintenance, and reproduction of research rodents. Autoclavable diets are supplemented with additional vitamins to compensate for losses that occur during autoclaving. Since our diets are nutritionally complete and balanced, it is not necessary to add dietary supplements.
There is no definitive point where one is able to predict when a specific diet will spoil or become deficient in one or more nutrients. The common guideline of a six month shelf life is based on longstanding practice in North America. In Europe and Asia, differences in local practices and regulatory oversight have led to Teklad standard natural ingredient diets being routinely used out to nine months and sometimes 12 months post-manufacture. This practical experience, along with literature support and vitamin testing over time, gives us confidence that these diets continue to support animal health and study integrity out to at least nine months post-manufacture. Please refer to your institution for guidance if you are unsure of local policies.
Recommended storage conditions:
- Cool and dry; at or below 70 degrees fahrenheit with humidity ideally below 50%, but up to 65% is acceptable
- Clean and free of pests
- In original packaging or in a container that prevents continuous exposure to light and minimal exposure to air
Standard natural ingredient rodent diets are complete diets for rats, mice, gerbils and hamsters, that should be allowed ad libitum access to the diet. Fresh water should be available at all times.

Description
StockOptions™ Polymer is a ready to use set of sterile filtered stock polymer solutions designed for the crystallization of biological macromolecules. StockOptions Polymer reagents are supplied in varying concentrations, each concentration appropriate for each particular polymer’s solubility and application as a primary or secondary precipitant, or additive in a crystallization experiment. StockOptions Polymer is comprised of 24 unique reagents.
StockOptions Polymer is designed to help researchers improve the speed, accuracy, precision, and quality of the formulation of crystallization screen reagents and crystallization optimization reagents. Researchers can use the individual StockOptions reagents to conveniently formulate custom screen solutions or accurately reproduce standard screen solutions from Hampton Research kits such as Crystal Screen™, Crystal Screen Cryo™, Crystal Screen Lite™, Natrix™, Crystal Screen 2™, Index™, PEGRx™ 1, PEGRx™ 2 and PEG/Ion™. StockOptions Polymer reagents can also be used to create solutions for the refinement and optimization of preliminary crystallization conditions. Finally, StockOptions Polymer reagents can be used to create accurate, precise, reproducible, high quality solutions for the production of single crystals.
StockOptions Polymer contains 24 reagents in sterile, screw cap polypropylene tubes. Each tube contains 10 milliliters of reagent and is purged with argon to promote reagent stability.
StockOptions Polymer contains Jeffamine® ED-2001 pH 7.0, Jeffamine® M-600 pH 7.0, Pentaerythritol ethoxylate (3/4 EO/OH), Pentaerythritol ethoxylate (15/4 EO/OH), Pentaerythritol propoxylate (5/4 PO/OH), Ethylene imine polymer, Poly(acrylic acid sodium salt) 5,100, Polyethylene glycol 200, Polyethylene glycol 300, Polyethylene glycol 400, Polyethylene glycol 600, Polyethylene glycol 1,000, Polyethylene glycol 1,500, Polyethylene glycol 3,350, Polyethylene glycol 4,000, Polyethylene glycol 6,000, Polyethylene glycol 8,000, Polyethylene glycol 10,000, Polyethylene glycol 20,000, Polyethylene glycol monomethyl ether 550, Polyethylene glycol monomethyl ether 2,000, Polyethylene glycol monomethyl ether 5,000, Polypropylene glycol P 400, and Polyvinylpyrrolidone K 15.

Double/Bubble® Epoxy HR4-347 extra fast setting with a work time of 3 to 5 minutes. The preferred epoxy for cryocrystallography. Sold as a pack of 5. Hardman Double/Bubble Fast-Setting Epoxy is an extra fast setting, two part, room temperature curing epoxy adhesive. Description
Micro-Tools™ are the smallest available precision instruments for laboratory use. These tools are realistically proportioned for macromolecular and small molecule crystallization work. The high quality, precision Micro-Tools ease microscopic manipulations. The eight specifically engineered Micro-Tools are useful in a variety of applications in the crystallization lab. Eight tools, a handle and wood storage box.
Micro-Tools are designed to be versatile as well as useful for specific crystallization manipulations. For example, the Micro-Scale is very useful for measuring crystal dimensions as well as serving as a way to record the dimensions by photography for reference or publication. The Micro-Chisel and Micro-Spade are helpful for separating and splitting blade and whisker clusters during seeding or mounting. The Micro-Prober and Micro-Needle come in handy when probing precipitate or when manipulating small or large crystals for seeding as well as during mounting. The Micro-Spatula can be used to move crystals as well as split single crystals or clusters. The Micro-Scraper is helpful when working with very small crystals.
The Micro-Tools Set is indispensable during crystal observation, manipulation, seeding, and mounting. The set includes a single anodized aluminum handle which holds each of the eight unique tips, all supplied in an attractive alderwood instrument case. The case provides protective storage of the Micro-Tools as well as a convenient carrying case.
The tips are constructed from hardened tungsten steel and are resistant to most common reagents used for crystal growth. The threaded tool tips are easily interchanged with the threaded aluminum handle. Then overall length of the handle only is 120 mm. The diameter of the tool shaft is 0.25 mm – 0.5 mm (0.01 – 0.02 inches). The actual dimension at the very tip of the tool will vary with the particular tool design.

Note: If you are using the CrystalCap, CrystalCap ALS, CrystalCap ALS HT, or CrystalCap SPINE HT, save time and money by using the ready to mount 18 mm Mounted CryoLoop.
Mounted CryoLoops with 20 micron diameter nylon. These nylon loops are bonded to hollow, stainless steel MicroTubes™ that are used to mount, freeze, and secure the crystal during cryocrystallographic procedures and X-ray data collection. The MicroTube is 24 mm in length and is specially engineered with EasySnap™ notches at the 10, 12, 14, 18, and 21 mm measures. To obtain the desired length of MicroTube, simply snap the MicroTube at the desired length and secure to the CrystalCap using epoxy or similar adhesive. These nylon loops show minimal diffraction, are thin for fast freezing, strong, and aerodynamic..
Most data collection geometries prefer or require a 22 mm length between the base of the cap and the beam. For a 22 mm overall length using the CrystalCap, CrystalCap ALS, CrystalCap ALS HT and CrystalCap SPINE HT, snap the MicroTube at the second notch from the bottom, creating an 18 mm MicroTube lenghth and secure to the CrystalCap using epoxy or similar adhesive. For a 22 mm over length using the CrystalCap Copper, CrystalCap Copper HT, CrystalCap ALS Copper HT snap the MicroTube at the fifth notch from the bottom (notch closest to the CryoLoop) and secure to the CrystalCap using epoxy or similar adhesive.

Vitamin adjusted diets

A few vitamin adjusted formulas are shown below. Please contact us for additional formulas of this nature or for more information about altering the vitamin profile of a diet, and please read below to learn more.
Most vitamin deficient diets use vitamin-free test casein (VFT casein) as the protein source. VFT casein is casein that has been extracted with denatured alcohol to reduce the fat content from about 1% to 0.1%. In the process the content of a number of vitamins (present at low or trace levels in casein) is further reduced, making this an ideal protein source for these types of diets. Envigo produces our own VFT in-house, and we also offer this as an ingredient for those mixing their own diets.
From this vitamin deficient base, various levels of vitamins can be added back. Many formulas adjust multiple vitamins, and other nutrients.
If use of an intact protein source is not advisable, an amino acid defined diet can be used.

Description
Used during cryocrystallography to displace and reduce the amount of water (mother liquor, reagent) on the crystal after the crystal is mounted in a cryoloop. Coating the crystal with oil can minimize evaporation from the crystal and reduce exposure and slow diffusion of air (oxygen) to the crystal.
Perfluoropolyether Cryo Oil offers low viscosity, extremely low vapor pressure, low surface tension, excellent chemical inertness and excellent thermal stability.
Dip the mounted crystal into the oil to displace surface water. Or add a small amount of Perfluoropolyether to the crystallization drop before mounting the crystal. After adding the Perfluoropolyether, mount the crystal using a Mounted Cryoloop. Withdraw the mounted crystal from the drop and the Perfluoropolyether will coat the mounted crystal. Cryogenically cool the mounted crystal.

Description
The Reductive Alkylation Kit offers a flexible alkylation protocol for methylation or ethylation or isopropylation of lysine residues.
Reductive alkylation of lysine residues to change protein properties (pI, solubility and hydropathy) which may promote crystallization via improved crystal packing.
Reductive alkylation of proteins has been successfully applied to obtain a significant number of high-quality crystals from proteins previously unable to be crystallized. Alkylating the e amino group of lysines alters the hydropathy, solubility and pI of the protein which may promote crystallization by altering sample-sample, sample-solvent and crystal packing interactions.
Reductive alkylation does not change the intrinsic charge on a protein but may change the isoelectric point (pI) slightly. The N-terminal amino group on the backbone will also be reductively alkylated. In general, alkylated proteins retain their original biochemical function. This protocol is designed with the goal of generating a high degree of modification with few side reactions, resulting in a homogeneous population of protein.

Description
The Seed Bead kits are used to create a seed stock for performing subsequent seeding experiments. The Seed Bead kits contains 24 Seed Bead Tubes, a special 1.5 ml microcentrifuge tube, shaped to maintain the seed in the sample during mixing. Each tube is configured with a PTFE, Stainless Steel, Zirconium Silicate Ceramic, or Glass Seed Beads.
Seeding allows one to grow crystals in the metastable zone. Crystallization in this zone provides control, reproducibility and an improved likelihood of a successful crystallization experiment. Also, crystals can grow from seeds but cannot spontaneously nucleate. By placing a seed or solution of seeds in a drop which is saturated to the metastable zone, one can use the seeds to grow larger single crystals. By controlling the number of seeds introduced into the drop, one can control the number of crystals grown. It is not practically possible to measure and know the number of seeds introduced to a drop, but by performing serial dilutions from a concentrated seed stock, one can control the number of crystals grown in the drop.
Using the Seed Bead kits, one can create crystal seed stock for subsequent seeding experiments. Crystals are placed in the microcentrifuge tube with the Seed Bead(s) and either vortexed or sonicated to generate a seed stock. Then by performing serial dilutions, one can control the number and size of crystals in the experiment.
The Seed Bead kit is useful for the preparation of seed stocks for automated and semi-automated microseeding (D’Arcy 2007, Harlos 2008).
Each Seed Bead Kit (HR2-320) contains 24 special microcentrifuge tubes, each tube containing a single 3mm PTFE Seed Bead.
Each Seed Bead Steel Kit (HR4-780) contains 24 special microcentrifuge tubes, each tube containing a six 1.6 mm Stainless Steel Seed Beads.
Each Seed Bead Ceramic Kit (HR4-781) contains 24 special microcentrifuge tubes, each tube containing a ten 1.0 mm Zirconium Silicate Ceramic Seed Beads.
Each Seed Bead Glass Kit (HR4-782) contains 24 special microcentrifuge tubes, each tube containing a ten 1.0 mm Glass Seed Beads.

Description
Grid Screen™ MPD is a preformulated reagent kit designed to provide a rapid screening method for the crystallization of biological macromolecules.
The screen is simple and practical for finding initial crystallization conditions as well as determining the solubility of a macromolecule in (+/-)-2-Methyl-2,4-pentanediol (MPD) between pH 4.0 and 9.0.
Grid Screen MPD evaluates (+/-)-2-Methyl-2,4-pentanediol (MPD) at four concentrations (10, 20, 40, 65% v/v) versus six pH levels (4, 5, 6, 7, 8, 9).
Grid Screen MPD consists of 24 unique reagents, each supplied as 10 milliliter in sterile, screw cap tubes. Ready-to-use reagents are sterile filtered and formulated with ultra-pure Type 1 water, using the highest purity polyol and buffers. Individual reagents are available through the Hampton Research Custom Shop.

Amino acid defined

A few amino acid defined formulas are shown below. This type of diet is used when a diet needs to be deficient in one or more amino acids, or adjustments need to be made to specific amino acid levels.
Formula examples:
Sometimes this type of diet is used to further reduce the background level of certain vitamins that are found in protein sources such as casein or vitamin-free (alcohol extracted) casein. Some folic acid deficient diets utilize this approach.
The amino acid profile for most amino acid defined diets is based on work by Harper and Rogers (J. Nutr. (1965) 87:267-276) and is not representative of the amino acid profile of an intact protein source such as casein. Other amino acid profiles can be specified. When an amino acid(s) is removed or reduced, isonitrogenous adjustment can be made if required.
