Description
PEG/Ion 400 is a crystallization reagent kit designed to provide a rapid screening method for the crystallization of membrane and soluble biological macromolecules.
Most G protein-coupled receptors (GPCRs) have been crystallized in Polyethylene glycol (PEG) 400 based reagents1. PEG/Ion 400 is designed as a 96 reagent crystallization screen that is compatible with the Lipidic Cubic Phase (LCP).
PEG/Ion 400 is supplied in a sterile, polypropylene 96 Deep Well block, each reservoir containing 1 ml of sterile filtered reagent. The block is compatible with robotic and multi-channel pipet liquid handling systems and is heat sealed using a special polypropylene backed film. Each PEG/Ion 400 is supplied with an adhesive sealing film which can be used to seal the block after removing the heat seal.
The screen combines a single concentration (30% v/v) of high purity Polyethylene glycol 400 and 48 different high purity salts, comprising both anions (malate, malonate, nitrate, phosphate, succinate, sulfate, tartrate, and thiocyanate) and cations (ammonium, cadmium, calcium, ethylammonium, lithium, magnesium, nickel, potassium, sodium, tetraethylammonium, and zinc) in two concentrations (0.1 and 0.4 M) which due to their unique pH characteristics also affords a reasonable pH screen (approximate pH range of 4 to 9.5). The primary screen variables are PEG, ion type, ionic strength, salt concentration, and pH.

Description
24 well crystallization plate for hanging drop or sitting drop vapor diffusion crystallization (when used with Micro-Bridges or Glass Rods), or dialysis crystallization (when used with Dialysis Buttons). Stackable, optically clear plastic 24 well plates with raised covers (to allow room for cover slides) and flat bottoms for exceptional optics. Raised, wide rings around each reservoir (well) minimize cross contamination and allow each well to be individually sealed with 22 mm diameter circle or square cover slides. Plates are individually wrapped and supplied without sealant.
15.0 cm x 10.8 cm plate footprint is convenient for manual pipetting and well access while at the same time compatible with some automated liquid handling systems.
Approximate dimensions: 15.0 cm x 10.6 cm x 2.2 cm.
Approximate well size: 1.7 cm x 1.6 cm.
Typical well volume: 500 to 1,000 µl.
Well capacity: 3.5 ml.

Description
As in selecting reagents for crystallization, the selection of a suitable cryoprotectant involves some trial and error as well as a screening.
A suitable cryoprotectant, when mixed with the crystal and crystallization reagent, will cool to cryogenic temperature without ice formation and damage to the crystal. To assay for the proper concentration of cryoprotectant in the reagent used to grow the crystal, one can mix the cryoprotectant with the crystallization reagent and employ the desired cooling method (for example, place the solution in a CryoLoop™ and place the CryoLoop in a cryostream). Observe for ice formation either visually or with x-ray diffraction. Upon cooling, a transparent drop and x-ray diffraction pattern, free of powder diffraction rings or “ice rings” indicates success. The appearance of a cloudy drop or “ice rings” indicates an inappropriate cryoprotectant concentration or cryoprotectant. Incrementally increase the concentration and/or composition of the cryoprotectant serially 5 to 10% and repeat until the cooled drop remains clear while in the cryostream. Once a clear drop is achieved in the cryostream, this is typically a good starting point for cryopreservation of the crystal.
Some crystals can simply be dipped or washed quickly in a simple cryoprotectant for successful cryopreservation. However, when all else fails, a rational assay of each cryoprotectant with incremental increases in cryoprotectant concentration as well as a test of mixtures can be help to identify the best cryoprotectant for a crystal.
Each CryoPro kit contains 1 milliliter each of 48 unique cryoprotectants. Supplied as one set of two boxes, each containing 24 reagents.
Ready to use cryoprotectants are formulated using high purity reagents with CryoPro reagents and are formulated using Type 1+ ultrapure water, 18.2 MO•cm resistivity at 25°C, < 5 ppb Total Organic Carbon, < 1 Bacteria (CFU/ml), < 0.03 Endotoxin (EU/ml) and are sterile filtered.

Amino acid defined
A few amino acid defined formulas are shown below. This type of diet is used when a diet needs to be deficient in one or more amino acids, or adjustments need to be made to specific amino acid levels.
Formula examples:
Sometimes this type of diet is used to further reduce the background level of certain vitamins that are found in protein sources such as casein or vitamin-free (alcohol extracted) casein. Some folic acid deficient diets utilize this approach.
The amino acid profile for most amino acid defined diets is based on work by Harper and Rogers (J. Nutr. (1965) 87:267-276) and is not representative of the amino acid profile of an intact protein source such as casein. Other amino acid profiles can be specified. When an amino acid(s) is removed or reduced, isonitrogenous adjustment can be made if required.

Description
MicroWick is useful for removing mother liquor from inside capillaries during crystal mounting. MicroWick is a non-metallic syringe needle used for filling and removing liquid from capillaries and micropipettes. MicroWick features a 67 mm tip, 0.164 mm OD, 0.1 mm ID (34 gauge) tip which will fit into most capillaries and micropipettes with an internal diameter of 0.2 mm or larger. The MicroWick needle is constructed from a combination of plastic and fused silica – no metal components are used. The MicroWick tip elasticity is sturdy and flexible though not unbreakable (much more flexible than glass!). Moderate bending will not block or damage the needle. The MicroWick luer fitting allows easy coupling to syringes and syringe filters. Each MicroWick contains one needle and one 1 ml syringe. Crystallization and capillary accessories sold separately.

Description
Individual reagents from the Proti-Ace and Proti-Ace 2 kits. Each protease is supplied in a stable, lyophilized format in an optimized digest buffer. Simply add water when ready to use.
The unique freeze dried formulation of the Proti-Ace kit offers a much improved protease stability compared to liquid protease formulations. Each tube contains 0.1 mg of lyophilized enzyme and digest buffer as shown below. Add 100 microliters of deionized water (Type 1+) to each tube of lyophilized content to create a 1 mg/ml protease solution in digest buffer. Proti-Ace Dilution buffer is supplied as a ready to use solution.
Proti-Ace Reagent 1* (Qty 3):
• 1 mg/ml a-Chymotrypsin, 1 mM Hydrochloric acid, 2 mM Calcium chloride dihydrate
Proti-Ace Reagent 2* (Qty 3):
• 1 mg/ml Trypsin, 1 mM Hydrochloric acid, 2 mM Calcium chloride dihydrate
Proti-Ace Reagent 3* (Qty 3):
• 1 mg/ml Elastase, 200 mM Tris pH 8.0
Proti-Ace Reagent 4* (Qty 3):
• 1 mg/ml Papain, 2 mM EDTA, 2 mM TCEP hydrochloride, 2 mM L-Cysteine
Proti-Ace Reagent 5* (Qty 3):
• 1 mg/ml Subtilisin, 10 mM Sodium acetate trihydrate pH 7.5, 5 mM Calcium acetate hydrate
Proti-Ace Reagent 6* (Qty 3):
• 1 mg/ml Endoproteinase Glu-C (in deionized water)
* When 100 µl of deionized water is added to each Proti-Ace freeze dried reagent
Proti-Ace Dilution Buffer (Qty 3):
• 1.6 ml of 10 mM HEPES pH 7.5, 500 mM Sodium chloride
Proti-Ace 2 Reagent 1* (Qty 3):

Description
StockOptions Buffer kits are reagent toolboxes for the macromolecular crystal grower. Stock-Options reagent kits offer precisely formulated, high quality crystallization grade reagent stocks in convenient, cost-effective kits. The chemicals utilized in StockOptions kits are the same crystallization grade, ultra-high purity chemicals utilized in the Hampton Research kits. StockOptions reagents are carefully formulated under strict quality standards to ensure reliable performance and lot-to-lot consistency.
Preformulated stocks remove calculation, measurement, and formulation errors. No more second guessing how the reagent was formulated, what specific chemical was used, when it was made, or how to precisely reproduce that reagent when it is gone and more reagent is required for additional setups.
StockOptions™ Sodium Citrate buffer kit is a preformulated, sterile filtered set of titrated buffer stocks. The StockOptions buffer stock reagents are supplied as 1.0 M stock solutions in 10 milliliter volumes. Each StockOptions Sodium Citrate buffer reagent is carefully titrated using hydrochloric acid. StockOptions Sodium Citrate is comprised of 24 unique reagents covering the pH range of 4.2 – 6.5 in 0.1 pH unit increments.
Formulated using Sodium citrate tribasic dihydrate and HCl.

Protein adjusted diets
A few protein adjusted formulas are shown below. Please contact us for additional formulas of this nature or for more information about altering the protein type or content of a diet.
Formula examples:
These diet examples are from a series of diets with the following features: casein-based, isocaloric (3.8 kcal/g) and matched for fat (5.5%), calcium (0.7%), and phosphorus (0.54%). There are other diets available from this series of diets, as well as other unique protein adjusted diets.
Casein is the most widely used refined protein source, but it is also possible to use other refined protein sources, such as “vitamin-free” test casein (alcohol extracted), lactalbumin, isolated soy protein, and egg white solids. These particular protein sources are usually used to achieve a specific research objective, rather than for routine use. For instance, vitamin-free test casein is most appropriately used in vitamin deficient diets, and egg white solids is most suitable for zinc or biotin deficient diets.
Adjusted protein diets usually range from “protein-free” (trace) to 60% protein, depending on the research.

To screen pH as a crystallization variable using Tacsimate one can mix together different ratios of different pH Tacsimate solutions. Refer to the pdf “Mixing Tacsimate Stocks to Screen pH” on this page. Or, one can add a buffer to the crystallizaiton reagent containing Tacsimate. Recommended final buffer concentration in the reagent well is 0.05 to 0.1 M.

A pH titrated mixture of organic acids. 1.8305 M Malonic acid, 0.25 M Ammonium citrate tribasic, 0.12 M Succinic acid, 0.3 M DL-Malic acid, 0.4 M Sodium acetate trihydrate, 0.5 M Sodium formate, 0.16 M Ammonium tartrate dibasic. pH adjusted using NaOH

The raw stock for our corncob bedding products is 100% corncob. No other components or additives are used. The 1/4″ (7097) and 1/8″ (7092) products are produced from only the woody-ring portion of the corn cob, and the pelleted corncob is produced from the pith and chaff (beeswing) portions.
As the raw stock corncobs are introduced to the plant, they pass through a hammermill that produces corncob fractions. The corncob fractions are then heat-dried. The drying process utilizes a rotary dryer systemthat heats the corncob fractions to approximately 200 degrees fahrenheit for two to three minutes. An aspiration process separates the woody-ring portion of the corncob from the pith and chaff portions. Smaller fractions/particles are produced by rollermill and hammermill processes. The particles are then sized and separated through a sifting process which utilizes screens conforming to U.S. standards. The particles pass through a final aspiration process to remove any remaining dust and aspiration process to remove any remaining dust and are then placed in overhead storage until packaged. The pith and chaff portions are processed through a pellet mill to produce the pelleted corncob product. Plant operating procedures require a minimum testing frequency of twice per shift for measuring moisture content and particle size distribution.

Description
Ionic Liquid Screen is a kit designed to allow the rapid and convenient evaluation of 24 unique ionic liquids and their ability to influence the crystallization of the sample. The screen is designed to be compatible with most crystallization reagents including all reagents utilized in all of the Hampton Research screens.
Ionic liquids have been found effective as additives in protein crystallization, with different ionic liquids used to increase crystallization rates and crystal size.1-4 The inclusion of ionic liquids in crystallization experiments has been reported to lead to less crystal polymorphism as well as less precipitation at higher precipitant concentrations.25 Ionic liquids have been used as additives to produce crystals in reagents that had previously not resulted in crystallization and results suggest ionic liquids may be applicable for the solubilization and crystallization of membrane proteins.2
Ionic liquids are organic salts with melting points below 100°Celsius. They are thermally stable, nonflammable and demonstrate very low vapor pressure. Ionic liquids are soluble in a variety of organic and inorganic reagents and can be highly water soluble. Ionic liquids can demonstrate a degree of localized structuring about each ion compared to materials composed of disassociated ions, setting them apart from salt solutions.5-6 Ionic liquids can participate in ionic, hydrophobic and hydrogen bond interactions. Ionic liquids are often chaotropic, composed of low symmetry ions with charge delocalization and weak intermolecular interactions.1 These organic salts generally consist of combinations of organic cations and either an organic or inorganic anion. Ionic liquids have been demonstrated to suppress protein aggregation and significantly increase protein folding yields.7-8 Ionic liquids have been reported to stabilize protein activity and structure.9-11 The inclusion of the ionic liquid 1-n-Butyl-3-methylimidazolium tetrafluoroborate (included in this kit) improved the thermal stability and solubility of integral membrane proteins for membrane proteomics study.12
Some ionic liquids, such as ethylammonium nitrate have water-like characteristics, including the capacity for hydrogen bonding and the promotion of micelle formation by some surfactants.13 Many ionic liquids are also organics acids and have ionic character in addition to the hydrophobic behavior, which makes them unique and useful solvents in protein chemistry.
Variation of the anion and the cation as well as the utilization of both soft (formate and acetate) and hard anions (nitrate) in the Ionic Liquid Screen reagents provides an additional dimension for evaluating the effects on ionic liquids on the solubility and crystallization of proteins. The Ionic Liquid Screen contains a set of 24 water soluble ionic liquids that comprise different cation (imidazolium, phosphonium, ammonium) and anion (borate, halides, sulfates, acetates, sulfonates, nitrates) structures for a diverse ionic liquid additive screen for use in improving the crystallization behavior and X-ray diffraction resolution of proteins.
Each of the 24 ionic liquids are preformulated in deionized water and sterile filtered using a 0.2 micron filter. Ionic liquid Screen contains 24 unique reagents, 0.5 milliliter each.
